Inward currents induced by ischemia in rat spinal cord dorsal horn neurons

چکیده
Hypoxia and ischemia occur in the spinal cord when blood vessels of the spinal cord are compressed under pathological conditions such as spinal stenosis, tumors, and traumatic spinal injury. Here by using spinal cord slice preparations and patch-clamp recordings we investigated the influence of an ischemia-simulating medium on dorsal horn neurons in deep lamina, a region that plays a significant role in sensory hypersensitivity and pathological pain. We found that the ischemia-simulating medium induced large inward currents in dorsal horn neurons recorded. The onset of the ischemia-induced inward currents was age-dependent, being onset earlier in older animals. Increases of sensory input by the stimulation of afferent fibers with electrical impulses or by capsaicin significantly speeded up the onset of the ischemia-induced inward currents. The ischemia-induced inward currents were abolished by the glutamate receptor antagonists CNQX (20 microM) and APV (50 microM). The ischemia-induced inward currents were also substantially inhibited by the glutamate transporter inhibitor TBOA (100 microM). Our results suggest that ischemia caused reversal operation of glutamate transporters, leading to the release of glutamate via glutamate transporters and the subsequent activation of glutamate receptors in the spinal dorsal horn neurons.

Comparison of Vulnerability by in Vitro Ischemia in Ventral and Dorsal Horn Neurons of the Rat Spinal Cord
INTRODUCTION Spinal Cord Injury (SCI) results in a loss of function such as mobility or feeling. SCI can be divided into two types of injury complete and incomplete. A complete injury means that there is no function below the level of the injury; no sensation and no voluntary movement. An incomplete injury means that there is some functioning below the primary level of the injury. A patient wit...
متن کاملLeukotriene enhances NMDA-induced inward currents in dorsal horn neurons of the rat spinal cord after peripheral nerve injury
BACKGROUND LTB4 is classified as a leukotriene (LT), a group of lipid mediators that are derived from arachidonic acid. It is recognized that leukotrienes are involved in the pathogenesis of many diseases, including peripheral inflammatory pain. However, little is known about the effects of leukotrienes on the spinal dorsal horn during neuropathic pain. Previously, we reported that there was in...
متن کاملRole of nitric oxide and Jun N-terminal kinase in the development of dark neurons in the dorsal horn of the spinal cord following induction of inflammatory pain
Introduction: Dark neurons which their morphological characteristics are consistent with those of cells undergoing apoptosis, are generated as an acute or delayed consequence of several pathological situations. The present study was designed to evaluate whether inflammatory pain regarding the role of NO and JNK lead to the formation of dark neurons in the dorsal horn of the lumbar spinal cor...
متن کاملThe activation of nicotinic acetylcholine receptors enhances the inhibitory synaptic transmission in the deep dorsal horn neurons of the adult rat spinal cord
Somatosensory information can be modulated by nicotinic acetylcholine receptors (nAChRs) in the superficial dorsal horn of the spinal cord. Nonetheless, the functional significance of nAChRs in the deep dorsal horn of adult animals remains unclear. Using whole-cell patch-clamp recordings from lamina V neurons in the adult rat spinal cord, we investigated whether the activation of nAChRs could m...
متن کاملJPET #208363 1 Nerve Injury Increases GluA2-lacking AMPA Receptor Prevalence in Spinal Cords: Functional Significance and Signaling Mechanisms
The glutamate AMPA receptors (AMPARs) are critically involved in the excitatory synaptic transmission, and blocking AMPARs at the spinal level reverses neuropathic pain. However, little is known about changes in the composition of synaptic AMPARs in the spinal dorsal horn after peripheral nerve injury. AMPARs lacking GluA2 subunit are permeable to Ca and their currents show unique inward rectif...
